In addition to histological analysis, the amounts of collagen in aorta were also measured biochemically by quantifying hydroxyproline, a non-proteinogenic amino acid mainly present in collagen, as described previously (Prockop and Udenfriend, 1960). Aortas were incubated in 6 M HCl at 95 °C for 20 h to hydrolyze collagen to free amino acids.

Liver collagen content was estimated by measuring the hydroxyproline content in fresh hepatic tissue with Ehrlich's reagent, according to a previously reported method (Prockop and Udenfriend, 1960). Liver samples were fixed with 4% paraformaldehyde (dissolved in 1X PBS) for 24 h. Tissue samples were then washed with water, dehydrated in alcohol and embedded in paraffin.
